titleOfInvention | Method for producing peptides using engineered inteins |
abstract | The present invention provides a method for producing peptides by recombinant means. This peptide is expressed as part of a fusion peptide comprising the target peptide and the engineered intein. The invention also provides engineered inteins, fusion proteins comprising the inteins, and DNA constructs encoding the fusion proteins. Thiol-induced cleavage of the fusion protein yields the carboxy-terminal α-thioester of the target peptide. The carboxy-terminal α-thioester can in principle react with any nucleophile, and thus this strategy allows a wide range of carboxy-terminal modifications such as chemical ligation, bioconjugation or amidation. . The engineered intein of the present invention is minimized in size and has a cysteine mutation at a position corresponding to position 3 of the Mxe GyrA intein (SEQ ID NO: 1) by alignment, thereby reducing the expression level of the fusion protein. Increased and isolated target peptides result in high yields, so the method of the present invention is suitable for production scale. |
