abstract | The present invention relates to biological methods for assaying peptidoglycan (PGN) in a sample, particularly a sample of glucose polymer. The PGN assay consists of a) treating a sample of glucose polymer by sonication, heating and / or alkalinization, and b) treating the treated sample or dilution thereof with exogenous TLR2 (Toll-like receptor 2). And a reporter gene that is directly dependent on a signal transduction pathway associated with TLR2, in contact with the expressing recombinant cell. The reporter gene encodes a protein whose activity can be measured with or without a colored protein or fluorescent protein, or a substrate, c) a signal derived from the reporter gene, and d) the amount of PGN and the reporter gene A standard curve of correlation with signal intensity is used to determine the amount of PGN in the sample. |
