A method for separating a biological sample and providing a downstream bioassay for the sample. (A) providing a composition comprising nucleic acid captured by magnetic particles in a first chamber of a cassette having a first chamber and a reaction chamber; and (b) an eluted nucleic acid sample. Elution of nucleic acids from particles in the first chamber, and (c) the first chamber at least partially disposed between the second chamber and the first chamber; Magnetically drawing the magnetic particles into a cavity of a valve member movably disposed within a valve housing having an opening communicating with the second chamber; and (d) the eluted nucleic acid into the reaction chamber. Transferring at least a portion of the sample; and (e) amplifying one or more target nucleic acid sequences by mixing the nucleic acid sample and a PCR reagent in the reaction chamber.
