abstract A ketoreductase polypeptide for the reduction of acetophenone is provided. Enzymes belonging to the ketoreductase or carbonyl reductase class (EC 1.1.1.184) are those of optically active alcohols from the corresponding pro-stereoisomeric ketone substrate and by stereospecific reduction of the corresponding racemic aldehyde substrate. Useful for synthesis. The present disclosure provides modified ketoreductase enzymes that have improved properties compared to naturally occurring wild-type ketoreductase enzymes. Also provided are polynucleotides that encode the modified ketoreductase enzyme, host cells capable of expressing the modified ketoreductase enzyme, and methods of synthesizing various chiral compounds using the modified ketoreductase enzyme.
