PROBLEM TO BE SOLVED: To provide a method for diagnosing a patient who can be resistant to treatment by either natural variation of SMO genotype or acquired mutation and resistance. An isolated nucleic acid molecule and probe having a mutation at amino acid position 473 that is at least 95% identical to a specific sequence and encodes a mutant SMO protein. Use of an isolated mutant SMO protein and an antibody conjugated with a cytotoxic agent that specifically binds to the mutant SMO protein. Furthermore, amplifying a nucleic acid corresponding to the carboxy terminus of the transmembrane domain 6 of SMO suspected of containing a mutation or a fragment thereof, and the electrophoretic mobility of the amplified nucleic acid to the corresponding wild-type SMO gene or A method of detecting a mutated SMO gene in a sample comprising comparing the electrophoretic mobility of the fragment.
