abstract | [Problem] To amplify nucleic acids contained in cells in a microchamber, preparation that can be easily and reliably filled with a nucleic acid amplification reaction reagent in a short time and then sealed with a sealing solution Provide a method. A flow path device including a micro chamber chip having a micro chamber capable of containing and holding cells formed on a surface thereof and a ceiling forming member for forming a flow path having a ceiling on the top is used. A method for amplifying nucleic acids contained in cells in the microchamber, (1) a step of feeding a cell suspension into the flow path so as to accommodate the cells in the microchamber; (2) a step of sending a nucleic acid amplification reaction reagent to the channel and at least filling the microchamber with the nucleic acid amplification reaction reagent; and (3) a sealing solution being fed to the channel to fill the microchamber. A method for preparing nucleic acid amplification, comprising a step of filling the flow path with a sealing liquid so that nucleic acid amplification reaction reagents do not communicate with each other.
