Provided are a chemiluminescent protein chip measuring method having time saving, economy, accuracy and convenience, and a reagent kit used therefor. A substrate slide glass of a protein chip includes at least one measurement subregion, and one of the measurement subregions measures a part of a serum sample. Within the measurement sub-region, set two measurement plaque areas and a row of control plaque areas, in which one measurement plaque area has measurement plaques formed by a specific antibody of fixed α-fetoprotein, and the other The measurement plaque area has measurement plaques formed by fixed lentil lectin, and the control plaque area has control plaques formed by fixed bovine serum albumin.
