abstract A fully humanized monoclonal antibody that specifically binds to Toll-like receptor 2 (TLR2) and antagonizes TLR2 function, but is substantially non-immunogenic in humans. An antibody or antigen thereof that binds to an epitope of human Toll-like receptor 2 with a KD of 3 × 10 −8 M or less and mediates antagonism of Toll-like receptor 2 independently of CD32 (Fcγ receptor II) Isolated, wherein the epitope is recognized by a reference antibody, the reference antibody comprising a heavy chain variable region comprising a specific amino acid sequence and a light chain variable region comprising another specific amino acid sequence Monoclonal antibody or antigen-binding site thereof. Also provided are nucleic acids encoding such antibodies and the use of antibodies in medicine, particularly the treatment of inflammation and autoimmune diseases mediated by Toll-like receptor 2 activity and signaling.
