Methods and compositions for producing high titer, substantially purified preparations of recombinant adeno-associated virus (rAAV) that can be used as vectors for gene delivery. A method for generating a population of rAAV particles comprising the following a) to d): Process. a) providing an AAV producer cell comprising: (i) one or more AAV packaging genes; (ii) a heterologous non-AAV polynucleotide flanked by at least one AAV inverted terminal repeat (ITR) rAAV provector; and (iii) AAV helper virus; b) incubating the producer cell provided in step a) under conditions allowing AAV replication; c) after incubation in step b) Lysing the producer cell to produce an AAV producer cell lysate; d) purifying the AAV producer cell lysate from step c); |
