Design of soluble full-length HIV-1GP41 trimer |
abstract |
Described herein is a soluble comprising a partial ectodomain and a cytoplasmic domain fused to the bacteriophage T4 small external capsid (Soc) protein and the bacteriophage T4 fibrin foldon domain (Fd). It is the HIV-1 retroviral transmembrane glycoprotein gp41 trimer (Soc-gp41M-Fd). The gp41 trimer with a pre-hairpin structure can be used as a candidate for understanding the mechanism of viral entry and for the development of HIV-1 vaccines, diagnostics and therapeutics. Other secondary embodiments of the gp41 protein comprising different variants are also disclosed. According to one embodiment, the gp41 trimer is further attached to a cell penetrating peptide (CPP). A method for producing the gp41 trimer is also disclosed. |
