The present invention provides methods for diagnosing immune-mediated diseases, such as autoimmune diseases, allergic or inflammatory diseases. Diagnosis is made by detection of haptoglobin 2 heterozygous or homozygous genotypes, or by detection and quantification of pre-haptoglobin 2 mRNA or protein. After diagnosis, the disease can be treated by decreasing the cell permeability that leads to increased transepithelial electrical resistance, for example by administering the antibody directly against single-chain zonulin, whereby epidermal growth factor Inhibits the receptor and inhibits proteinase activated receptor 2 (PAR 2 ).
