abstract | A method for detecting a target nucleic acid sequence in a sample using a cycling probe reaction is provided. A) (i) an oligonucleotide probe having a cleavage domain lacking an RNA residue, (ii) a nucleic acid sample that may or may not have a target sequence, (iii) Providing a reaction mixture comprising a thermophilic RNase H2 enzyme capable of cleaving oligonucleotide probes when hybridized to a target nucleic acid sequence; b) probing the target DNA sequence to form a double-stranded substrate C) cleaving the hybridized probe using the RNase H2 enzyme at or within the cleavage domain or adjacent to the cleavage domain; and d) detecting probe cleavage.
