A method, machine, computer system, computer readable medium, and kit for predicting the effectiveness of a topoisomerase I (topo I) inhibitor in the treatment of cancer. A method, machine, computer system, computer readable medium, and kit that can be used to determine phosphorylation at residue serine 10 (S10) of a polypeptide of topoisomerase I polypeptide. Phosphorylation at serine 10 indicates that the cancer may be non-reactive with the topo I inhibitor, while the absence of phosphorylation at residue serine 10 (S10) indicates that the cancer is a topo I inhibitor.
