PROBLEM TO BE SOLVED To provide glycosylation of a molecule. Methods and genetically engineered cells useful for producing modified N-glycosylated forms of target molecules are described herein. Also disclosed are methods and molecules involving modified N-glycosylation useful for the treatment of various diseases such as metabolic disorders. In the present invention, a single gene deletion (outer chain extension (OCH1) deletion) in Yarrowia lipolytica cells has an α-1,2-linked mannose residue on the Man5GlcNAc2 (Structure Formula IV; FIG. 1) backbone Based on the finding that it resulted in substantially uniform production of glycosylated proteins.
