titleOfInvention | Ultra-concentrated fast-acting insulin analogue preparation |
abstract | A pharmaceutical formulation has a mutant insulin B-chain polypeptide comprising ortho-monofluoro-phenylalanine at position B24 in combination with an amino acid substitution containing an acidic side chain at position B10, wherein the insulin is 0.6 mM. It can be present at a concentration of ~ 3.0 mM. The formulation may optionally be free of zinc. In addition, one or more amino acid substitutions may be present at positions B3, B28, and B29. The mutant B chain polypeptide may be part of a proinsulin analog or a short chain insulin analog. The insulin analog can also be an analog of mammalian insulin such as human insulin. A method for reducing a patient's blood glucose level comprises administering to the patient a physiologically effective amount of the insulin analog or a physiologically acceptable salt thereof.
