abstract | The present invention provides a composition comprising a phosphorus-containing tricyclic compound comprising a phthalazin-1 (2H) -one derivative. These compounds are potent inhibitors of the enzyme poly (ADP-ribose) polymerase (PARP), in particular PARP-1, and potentially potent inhibitors of PARP-2. They also show good cellular activity in inhibiting poly (ADP-ribose) oligomer formation. These compounds may be useful in the treatment of conditions involving PARP, such as cancer, inflammatory diseases and ischemic conditions, as monotherapy or in combination therapy with other therapeutic agents. Accordingly, there is also provided a method for the treatment of a condition involving PARP comprising the step of administering an effective amount of a compound of the invention to an individual in need thereof. |
