abstract |
The present invention provides a method for obtaining a nucleic acid encoding an immunoglobulin variable domain from a single immunoglobulin-producing cell. A first polymerase chain reaction using 3 to 6 5 ′ primers and one 3 ′ primer, and 13 to 16 5 ′ primers and one 3 ′ primer are used. A method comprising a step of performing a second polymerase chain reaction with a product of the first polymerase chain reaction, wherein a distance between binding positions of primers used in the second polymerase chain reaction is reduced as compared with the first polymerase chain reaction.
