Provided are antibodies against factor Xa inhibitors and methods of use thereof. One of the major limitations of anticoagulant therapy is the risk of bleeding associated with therapy and the ability to rapidly reverse anticoagulant activity in the event of overdose or emergency surgical treatment This is a limitation. Therefore, specific and effective antidote for all forms of anticoagulant therapy is highly desirable. The present invention relates to an antidote to an anticoagulant that targets factor Xa. An antidote is a factor Xa protein derivative that binds to and substantially neutralizes a factor Xa inhibitor but does not aggregate to form a prothrombinase complex. The derivatives described herein lack or have a reduced intrinsic clotting activity. Disclosed herein are methods for stopping or preventing bleeding in patients currently undergoing anticoagulation therapy with a factor Xa inhibitor. [Selection figure] None |
