A novel enzyme for degrading isomaltose into glucose even under conditions of high glucose concentration, and a method for industrial production of glucose from starch. In an isomaltase having a specific amino acid sequence, or an isomaltase in which one to several amino acid residues are deleted, substituted or inserted in the amino acid sequence, one or more of amino acids of amino acid numbers 200 to 300 Isomaltase in which two or more are mutated to an amino acid selected from isoleucine, tryptophan and glutamic acid.
