titleOfInvention | Methods and compositions for quantitative detection of nucleic acid sequences over an extended dynamic range |
abstract | Methods and compositions for quantitative detection of nucleic acid sequences over an extended dynamic range are provided. Disclosed are compositions and methods for making equal ratios of distinguishable amplicon species using a single amplification system in a single tank. The number of distinguishable amplicons and their ratio to each other are such that they span the linear dynamic range required for the amplification reaction and of the measurement system used to determine the amount of amplicon produced. Selected to correspond to the limit. Providing non-uniform amounts of one or more amplification reaction components (eg, distinct amplification oligomers, natural and non-natural NTPs in NTP mixtures, etc.) produces non-uniform amounts of distinguishable amplicon species Is done. The amount of target nucleic acid in the test sample is determined using a linear detection range obtained from detection of one or more amplicon species having an amount within the dynamic range of detection.
