abstract The present invention provides detection and quantification of biomolecules using mass spectrometry. The present invention relates, in part, to a method for detecting a target nucleic acid using a detector oligonucleotide detectable by mass spectrometry. This method uses the 5 ′ → 3 ′ nuclease activity of a nucleic acid polymerase to cleave the annealed oligonucleotide probe from the hybridized duplex and release the label for detection by mass spectrometry. This process can be easily integrated into a PCR amplification assay. This method also includes embodiments relating to quantitative analysis of target nucleic acids. [Selection figure] None
