abstract | A method is provided for purifying N-methyliminodiacetic acid (MIDA) boronate from solution. Also provided are methods for deprotecting boronic acids from their MIDA ligands. Purification and deprotection methods can be used in combination with methods for coupling or otherwise reacting with boronic acids. Repeated cycles of deprotection, coupling, and purification can be performed to synthesize the compound of interest. The method is suitable for use in automated chemical synthesis processes. Also provided is an automated small molecule synthesizer for performing automated synthesis of small molecules using iterative cycles of deprotection, coupling, and purification according to the methods of the present invention. Coupling and other reactions included in the present invention include, without limitation, Suzuki-Miyaura coupling, oxidation, swern oxidation, “Jones reagent” oxidation, reduction, Evans Aldol reaction, HWE olefination, Takai olefination, Examples include alcohol silylation, desilylation, p-methoxybenzylation, iodination, leekcyclos coupling, Heck coupling, Miyaura labrillation, Stille coupling, and Sonogashira coupling. |
