Synthesis and use of kinase inhibitors |
abstract |
An improved synthesis of the class of adhesion plaque kinase (FAK) inhibitors is provided, where the use of expensive palladium-based catalysts is reduced and the reaction yield and product purity are improved. The two important reactions of aryl halide and aniline coupling are optimized by the surprising discovery that palladium-based catalysts can be eliminated entirely in one of the reactions. The invention also provides the use of FAK inhibitory compounds in the treatment of inflammatory and immune disorders, and arthritis. |
