abstract | An improved bacterial secretion signal derived from pelB and ompA is provided. Improved mutants enhance bacterial membrane secretion and thus proteins displayed on filamentous phage particles, and in particular in the polypeptide chain or in order to form correctly folded, functional protein structures. It is useful for the production of proteins secreted from bacteria, including proteins that require oxidative formation of covalent bonds such as disulfide bonds between polypeptide chains. A method for multivalent display of a combined synthetic library of such proteins presented as a fusion polypeptide with complex dimeric proteins on the surface of bacteriophage particles and filamentous phage pIX coat proteins As described herein. Heterodimers or more complex interchain linkage structures can also be presented using the methods of the invention. |
