Identification of agents capable of modulating human taste responses (particularly sweetness enhancers) by novel sweet taste receptor protein-based assays, heterologous expression systems comprising nucleic acid constructs forming said sweet taste receptor proteins, and Use of sweet receptor protein in screening is provided. A method for enhancing sweetness comprising using a sweetener in combination with other sweeteners at a threshold-threshold concentration at a concentration from sub-threshold to near-threshold.
