Provided is a binding molecule that specifically binds to GITR, eg, human GITR (hGITR), on T cells and dendritic cells. A binding molecule that binds to hGITR with high affinity in the presence of a stimulant, such as CD3, is agonistic and reverses the suppression of Teff cells by Treg cells. Various aspects relate to binding molecules, and pharmaceutical compositions thereof, and nucleic acids, recombinant expression vectors, and host cells for making such binding molecules.
