abstract A recognition domain and a cleavage domain of a FokI restriction endonuclease are identified, and a DNA segment encoding each of the recognition domain and the cleavage domain of the FokI restriction endonuclease is provided. The 41 kDa N-terminal fragment constitutes the recognition domain of the FokI restriction endonuclease, and the 41 kDa C-terminal fragment constitutes the nuclease domain of the FokI restriction endonuclease. A hybrid restriction enzyme comprising a nuclease domain of a FokI restriction endonuclease linked to the recognition domain of another enzyme. One such hybrid restriction enzyme is Ubx-F N. The enzyme contains a Ubx homeodomain linked to a FokI cleavage or nuclease domain. Two insertion variants of FokI endonuclease. [Selection figure] None
