A non-cytotoxic protease or fragment capable of cleaving a protein of an exocytosis fusion organ of a nociceptive sensory afferent nerve cell; A targeting moiety capable of binding to a binding site on the nociceptive sensory afferent neuron; a protease cleavage site located between the non-cytotoxic protease or fragment and the targeting moiety; and from within an endosome A single chain polypeptide fusion protein comprising a translocation domain capable of translocating the protease or fragment across the endosomal membrane into the cytosol of the nociceptive sensory afferents.
