abstract |
An analysis method using an unnatural base is provided. A sample presumed to contain a target nucleic acid is brought into contact with a nucleic acid polymerase and first and second primers; and has a double-stranded region 122 and a single-stranded region containing an unnatural base 114. Generating an amplification product 120; contacting the sample with a reporter 126 comprising a label 132 and a non-natural base 130 complementary to the non-natural base of the single-stranded region; at least a portion of the reporter being a single strand of amplification product Annealing to the region; after annealing, cutting at least a portion of the reporter to release at least one reporter fragment 134; and correlating the release of at least one reporter fragment with the presence of the target nucleic acid in the sample.
