abstract | The present invention provides acetal derivatives of luciferin and methods for using such derivatives in assays for enzyme activity. A derivative of luciferin acts as a substrate for the desired enzyme and is a pro-substrate for luciferase (proluciferin). In this way, a specific enzyme recognition site for an enzyme that is not the desired luciferase (an intramolecular chemical reactive group referred to as a substrate for this enzyme) is the skeleton of a luciferin derivative (or an appropriate luciferase for the luciferase). By providing a luciferin derivative linked to the other chemical moiety that constitutes the substrate, an enzyme that is not a luciferase can be measured in a bioluminescent assay. Such a derivative is, for example, a derivative having a modification at the carboxyl group of D-luciferin or aminoluciferin, and having other modifications as necessary. |
