The present invention provides a production and screening of a novel protease that cleaves a protein known to be involved in a disease. A method of identifying a protease that cleaves a substrate sequence, the method comprising generating a library of protease sequences, wherein each member of the library is against a wild-type backbone sequence. Having a protease backbone with N mutations; measuring the activity of each member of the library in cleaving the substrate sequence; and the activity of each member relative to the average activity of the library A method comprising the steps of comparing, thereby identifying which protease has the highest cleavage activity, wherein N is a positive integer.
