abstract |
An object of the present invention is to provide a screening method for a bone mass increasing agent, characterized in that the evaluation is performed using the degradation property of a vitamin D receptor protein by binding of a test substance as an index. In order to solve the above problems, the present inventors examined the amount of VDR protein in cultured cells derived from wild-type mice and human osteoblasts administered with ED-71. As a result, it was found that the amount of VDR protein was significantly reduced. It was also clarified that this proteolysis is caused by a calpain degradation system, which is a calcium-dependent cysteine protease. Furthermore, it was clarified that VDR to which ED-71 binds interacts with calpain regulatory subunit s1 and is actively degraded. [Selection figure] None |
