Provided is an antibody that is also known as ADDL, and recognizes and recognizes a multidimensional structure of an Aβ-derived diffusible ligand. Alzheimer's disease and control human brain extracts can be distinguished, which is useful in methods of detecting ADDLs and diagnosing Alzheimer's disease. The antibodies are also useful in methods for blocking ADDL binding to neurons, ADDL assembly and tau phosphorylation and thus preventing and treating diseases associated with soluble oligomers of amyloid β1-42.
