PROBLEM TO BE SOLVED: To provide a method for detecting a mutation having excellent detection sensitivity using Tm analysis. A sample containing a detection target DNA in which a detection site is mutated and a non-detection target DNA in which the detection site is unmutated is complementary to a detection target sequence containing the mutated detection site. A detection probe composed of a polynucleotide and a non-detection target polynucleotide containing an unmutated detection site containing the detection site are added, and the detection probe is hybridized to the DNA. Then, the presence or absence of mutation is determined by heating the hybrid of the DNA and the detection probe to measure signal fluctuations accompanying temperature rise, and analyzing the signal fluctuations to determine the Tm value.
