Provided herein are methods and compositions for amplifying nucleic acids. These methods involve the use of 3 'substituted nucleoside 5'-triphosphates or 3' substituted termination primers in nucleic acid amplification reactions. In certain embodiments, the method is accomplished by using 3 'substituted NTPs and / or 3' substituted termination primers that provide utility in nucleic acid amplification. In a preferred embodiment, the NTP and / or primer is substituted at the 3 'position with certain thermally labile chemical groups such as ethers, esters, or carbonates. |
