abstract | We disclose a particle comprising a matrix coated thereon and having a positive charge, the size of which allows for the assembly of primate or human stem cells attached thereto. The particles can include substantially elongated, cylindrical or rod-shaped particles having a longest dimension of 50 μm to 400 μm, for example about 200 μm. It can have a cross-sectional dimension of 20 μm to 30 μm. The particles can include substantially compact or spherical shaped particles having a size of about 20 μm to about 120 μm, for example about 65 μm. The present inventors provide a method for propagating primate or human stem cells, comprising preparing first and second primate or human stem cells attached to respective first and second particles, Contacting a primate or human stem cell with a second primate or human stem cell to form a cell aggregate, and culturing the aggregate to propagate at least one passage of the primate or human stem cell. A method of including is also disclosed. Also disclosed is a method of propagating human embryonic stem cells (hESC) in long-term suspension culture using microcarriers coated in Matrigel or hyaluronic acid.
