abstract The present invention relates to an in vitro method for detecting different age SGs in a cell by distinguishing and labeling the SGs according to the age of the secretory granules (SG) of different age. And the following steps: (A) A SG-specific (poly) peptide that has a binding site common to at least three different substances A, B and C that form SG and are covalently bound to it: 2. (i) comprising a (poly) peptide specific for SG, and (ii) a (poly) peptide having a binding site common to at least three different substances A, substance B and substance C covalently bound thereto. A cell capable of expressing a fusion protein; wherein said substance is capable of penetrating the cell membrane, and at least substance A and substance C are detectable by separate means, targeting a binding site Contacting with substance A; (B) a step of saturating the binding site by contacting the cell with a substance B that blocks a binding site that is not bound by the substance A; (C) allowing unbound substance B to be removed from the cell, or removing unbound substance B from the cell; (D) contacting the cell with a substance C that targets a binding site on a (poly) peptide or fusion protein and making it available for binding after step (b); and (e) detectable Detecting for the presence of both substance A and substance C; [where simultaneous detection of both substance A and substance C indicates the presence of two populations of SG with different ages] Relating to said in vitro method.
