abstract |
An improved system for nucleic acid cloning is provided. The present invention provides an improved system for linking nucleic acids to each other. In particular, the present invention provides techniques for generating DNA product molecules that can be easily and directly linked to recipient molecules. The present invention further provides a method for directed linking of product molecules (ie, selective linking of specific molecules within a collection of molecules). The present invention also provides a method of exon shuffling, wherein multiple different product molecules are produced in a single ligation reaction. The DNA manipulation system of the present invention can be easily incorporated into other nucleic acid manipulation systems (eg, ribozyme-mediated systems) and can also be automated.
