abstract The present invention provides a novel method for sequencing nucleic acids (eg, DNA), as well as improved methods for performing direct sequencing of nucleic acids (eg, DNA). At least one primer is annealed to a template polynucleotide sequence and the primer is extended in the presence of one or more nucleoside triphosphates to produce a modified polynucleotide sequence. At least one of the nucleoside triphosphates has been modified and the modified nucleotide sequence comprises a phosphorothiolate linkage. A modified polynucleotide sequence comprising a 3 ′ phosphorothiolate linkage or a modified polynucleotide sequence comprising a 5 ′ phosphorothiolate linkage is generated. [Selection figure] None
