Cell lines for expressing enzymes useful for the preparation of amidation products |
abstract |
A cell line for expressing peptidylglycine α-amidated monooxygenase (PAM), or one of its two catalytic domains. High levels of enzyme expression are achieved using low protein tissue culture media from non-animal sources. Robust two-step downstream purification results in high enzyme purity. The resulting PAM or its PHM catalytic domain is X-α-hydroxy-Gly or X-NH 2, where X is a peptide or any chemical compound having a carbonyl group to which a glycine group can be covalently bonded. -Used to catalyze enzymatic conversion of Gly conversion. A preferred cell line preparation method is also shown. [Selection] Figure 1 |
