Provided is a nucleic acid useful as a tool for triggering cell lethality of a tumor treated with anticancer treatment. A nucleic acid molecule comprising a double-stranded portion of 24-100 bp, having at least one free end, and a substrate for binding by at least a Ku protein involved in the NHEJ pathway of double-strand break repair. Preferably, it is a linear or hairpin nucleic acid molecule consisting of a DNA fragment, and the loop is a linker, hexaethylene glycol or tetradeoxythymidylate. Such double-stranded nucleic acid fragments can interfere with DNA damage signaling and repair pathways, particularly the heterologous NHEJ pathway of double-strand break repair, and are used in conjunction with DNA breakage treatments, particularly radiation therapy or chemotherapy.
