abstract | A method for suppressing a nonspecific reaction in detection of a target nucleic acid using a primer-immobilized substrate is provided. A method for detecting a target nucleic acid in a biological sample, wherein (1) the target nucleic acid does not contain a target nucleic acid and a labeled substrate and contains an unlabeled substrate on a substrate on which a primer is immobilized. A step of adding a reaction solution and a nucleic acid chain extending enzyme and pretreating the substrate at a temperature at which the enzyme functions; (2) containing a target nucleic acid, a labeled substrate and an unlabeled substrate on the substrate after the pretreatment; A step of adding a nucleic acid amplification reaction solution and a nucleic acid chain extending enzyme, and performing solid phase nucleic acid synthesis by an extension reaction of an immobilized primer using the target nucleic acid as a template at a temperature at which the enzyme functions, and (3 ) Detecting a synthetic strand of the target nucleic acid.
