abstract | A target sequence is expressed in a plant, a plant part, or a plant cell. A method of expressing a target sequence in a plant, plant part, or plant cell culture comprising: (a) a sequence encoding an RNA replicon operably linked to or capable of being linked to a transcriptional promoter. Preparing a plant, plant part, or plant cell culture containing heterologous DNA in the cell nucleus, wherein the sequence encoding the RNA replicon is (i) an RNA replicon derived from the sequence of a plant RNA virus Contains a sequence related to the replicon function, (ii) a target sequence, whereby the sequence related to the replicon function exhibits a function-conservative difference from the sequence of the plant RNA virus at a selected position of the sequence of the plant RNA virus. And the difference increases the frequency of replicon formation compared to an RNA replicon that does not exhibit the difference; and ( ) The method comprising expressing a sequence of interest.
