abstract | The present invention encompasses a model-based method for determining insulin sensitivity and glucose absorption from an oral glucose tolerance test or mixed diet. The present invention has several advantages over current approaches. The method requires about 4-6 blood samples taken about 2-3 hours after glucose intake and can therefore be applied to large clinical trials. This analysis method uses a simplified version of the classic minimal model, a method that expresses glucose absorption using only two parameters, and an integral method that makes it possible to obtain parameters using simple algebraic calculations. The method clearly identifies differences in insulin sensitivity in different patient types and the improvement in insulin sensitivity caused by drug therapy. Furthermore, the measured insulin sensitivity obtained by this method is highly correlated with the result obtained from the high insulin clamp method (r 2 > 0.8). Therefore, this method is a practical and reliable method for determining insulin sensitivity under physiological conditions. |
