abstract | a method for screening for a molecule that reduces apoptosis of a cell comprising: i) combining a candidate molecule with an assay cell; and ii) determining a change in the survival of the assay cell in the presence of the molecule relative to a control. I will provide a. In one embodiment, the assay cells are treated with an apoptosis inducing agent prior to steps i) and ii) or during steps i) and ii). In one example, the treatment agent decreases the level or activity of a pro-survival member of the Bcl-2 protein family such as Bcl x L or Mcl1. In another embodiment, the level or activity of at least one pro-survival member of the Bcl-2 family is reduced in the cells of step i). In some embodiments, this is independent of any effect of the candidate molecule or pro-apoptotic agent. By reducing the level or activity of one or more pro-survival Bcl-2 protein family members in an assay cell, the cell is mediated by, inter alia, Bak or Bax, or Bak and Bax, unless rescued by a candidate molecule. Will undergo apoptosis. |
