A method of providing a droplet having a reduced amount of material in contact with a magnetically responsive bead. The method generally includes (a) (i) a substrate including an electrode disposed to perform a droplet operation on a surface; (ii) (1) one or more magnetically responsive beads; Providing a droplet actuator comprising: (2) a starting amount of material; and (3) a starting droplet comprising a starting deposition; (b) one or more at a location remote from the target droplet splitting region (C) splitting the mixed droplets and (i) containing substantially all of the one or more magnetically responsive beads, reduced compared to the starting concentration One or more droplet operations comprising a droplet splitting operation selected to produce a droplet set comprising droplets having a reduced amount of material and (ii) droplets substantially lacking magnetically responsive beads The process of performing is included.
