I-CreI has at least one substitution in one of the two functional subdomains of the LAGLIDADG core domain located at positions 26-40 and 44-77 and has the nucleotide sequence of SEQ ID NOs: 1-14 An I-CreI mutant or single chain derivative capable of cleaving a DNA target sequence from the HPRT gene. Use of said variants for therapeutic (Resh-Nyhan syndrome gene therapy) or non-therapeutic purposes (engineering of transgenic animals and recombinant cell lines) to induce site-specific modifications in the HPRT gene .
