abstract [PROBLEMS] To detect and quantify a small number of microorganisms contained in a large amount of sample quickly, with high accuracy, with high sensitivity, and simply. An upper first layer 116 is used as a hydrophilic membrane filter, and a hydrophobic membrane filter 117 capable of filtering an aqueous solution by generating a negative pressure without using a wetting agent is provided below the second layer. Using a sample container 102 having a two-layer membrane filter 101 at the bottom, a large amount of sample aqueous solution is filtered by the negative pressure generated by the suction unit 105, and microorganisms in the sample aqueous solution are captured by the hydrophilic membrane filter. Further, after the negative pressure is changed to normal pressure, the microbial solution is added, and the microbial solution is retained on the hydrophobic membrane filter for a certain time.
