abstract |
Disclosed is a modified polysaccharide (especially a modified Neisseria meningitidis serotype A polysaccharide) that retains immunogenicity but has improved stability. Modified polysaccharides (especially modified Neisseria meningitidis serotype A polysaccharide) that retain immunogenicity but have improved stability. Typically, modified polysaccharides are prepared by reacting capsular polysaccharides or oligosaccharide fragments thereof with a bifunctional reagent such as CDI followed by an amino compound such as dimethylamine. Modified polysaccharide-protein conjugates and vaccines prepared from such conjugates have also been described. The present invention provides a modified capsular saccharide comprising a protecting group at the position of the hydroxyl group of at least one monosaccharide unit of the corresponding natural capsular saccharide.
