titleOfInvention | Method for detecting nucleic acid amplification reaction |
abstract | An object of the present invention is to provide a permanent and unaffected by the sequence of a sample nucleic acid without the addition of new reagents that may affect the amplification reaction or a special sensor electrode having a complicated configuration. It is to provide an accurate detection method. The present invention includes a step of adding a sample nucleic acid to a nucleic acid amplification buffer containing a reducing agent molecule, a redox molecule, and a magnesium ion and performing an amplification reaction; and the amplification reaction of the sample nucleic acid is carried out in the buffer. In the buffer, the pyrophosphoric acid produced with the amplification of the sample nucleic acid forms the magnesium ion and magnesium pyrophosphate, thereby reducing the concentration of the magnesium ion in the buffer. Detection of a nucleic acid amplification reaction, comprising: a step of measuring a reduction current generated by a reduction reaction between a molecule and the redox molecule; and a step of determining the presence or absence of amplification of the sample nucleic acid from the value of the measured reduction current Provide a method.
